Tell Me, O LJ Genie

So, at this workshop, we critique each others' first 50 pages of a novel, and then we break into smaller groups to critique the full books. And it's come up in the workshops a lot, enough that I've lost all objectivity on the issue, so I would like to put the question to you:

Do you need to know where the overall thrust of the book is going within the first 50 pages? Do you need to have a firm grip the central conflict before 50 pages are up? How long to you give a book to present its plot?
